phosphorylase kinase

phosphorylase kinase
phos·phor·y·lase ki·nase (fos-forґə-lās kiґnās) [EC 2.7.11.19] an enzyme of the transferase class that catalyzes the phosphorylation of (inactive) glycogen phosphorylase b to form (active) glycogen phosphorylase a, a step in the cascade of reactions regulating glycogenolysis. The enzyme is itself activated via phosphorylation by cyclic-AMP–dependent protein kinase. There are liver and muscle isoforms, each of which comprises four subunits, alpha, beta, gamma, and delta, encoded by multiple genes on multiple chromosomes; the alpha and gamma subunits are differentially encoded and expressed in different tissues while the beta remains constant across the isoforms and the delta is calmodulin. Deficiency of the enzyme causes glycogen storage disease, type IX (q.v.), which is divided into subtypes on the basis of which isoform and specific subunit the mutation affects. Written also phosphorylase b kinase.
