ligase chain reaction
a type of DNA amplification technique that uses DNA ligase to link adjacent pairs of synthetic oligonucleotides that have been hybridized to complementary regions on each strand of the double-stranded target DNA, with the product used as a template for subsequent cycles of amplification of the sequence in question. Because the ligation reaction requires perfect base pairing, sequence variation at the ligation site can be demonstrated.
