Lцffler alkaline methylene blue stain
a simple stain used especially for demonstrating granules in Corynebacterium diphtheriae. Methylene blue made alkaline with potassium hydroxide is applied to a smear briefly and the slide washed. Granules appear deep blue in lighter blue cells.
